Overview: 

Vanilla acetone is a synthetic raw material for a variety of psychotropic drugs and a variety of new antimalarial drugs, and is often used as a cardiotonic and diuretic in medicine. As a specific nicotinamide adenine dinucleotide oxidase inhibitor, nicotinamide adenine dinucleotide phosphate (NADPH) oxidase and xanthine oxidase are the main sources of free radicals during myocardial reperfusion. Therefore, inhibiting their activity and reducing the generation of free radicals may alleviate reperfusion injury. In this study, the use of the NADPH oxidase inhibitor vanilla acetone can exert a protective effect on the myocardium of reperfusion rats, which is related to the inhibition of the activity of NADPH oxidase and the reduction of its mediated generation of superoxide anion and nitrite peroxide free radicals.

Application: 

Vanilla acetone can be used in the fragrance industry is also an important organic synthetic raw material, can be used for the synthesis of psychotropic drug iperidone and a variety of new antimalarial drugs. It is also a commonly used antioxidant, is currently recognized as a specific nicotinamide adenine dinucleotide oxidase inhibitor, in medicine is also commonly used as a cardiac and diuretic.

Preparation: 

Anhydrous ethanol 25 ml, guaiacol 2.0 ml (18.0mmol), anhydrous potassium carbonate 5.0 g(36.2 mmol) sodium iodide 0.3 g(2.0 mmol) and propane bromide 3.2 ml(35.4 mmol) were added to the reaction bottle in sequence and stirred for reflux reaction for 10 h. Ethanol and excess propane bromide were removed by steam under filter and vacuum and added into 40 ml dichloromethane and 30 ml 1 mol-L-1 sodium hydroxide solution, washed in organic phase water (2x25 ml), dried with anhydrous magnesium sulfate, and yellow liquid 12.86g was obtained by spin evaporation with a yield of 95.3%. 25 ml and 11.5 g(9.0 mmol) of dichloromethane were added into the reaction bottle, and then 2.5 g(18.3 mmol) of anhydrous zinc chloride was added into the ice bath for cooling, and the mixed solution [dichloromethane (2.0 ml)+ acetic anhydride (1.7 ml,18.0 mmol)] was added. After drip, the reaction was slowly raised to room temperature for 4 h. 

40 ml 1 mol-L-1 hydrochloric acid was added, the aqueous phase was extracted with dichloromethane ( 2*30 ml), the extraction liquid was combined, washed with water (2×30 ml), and the yellow liquid mixture of 1.84 g was obtained by drying and spin evaporation of anhydrous magnesium sulfate with 97.9% yield and 99.1% purity. 16 mL of dichloromethane and the mixture of isomer 3-methoxy4-n-propoxyacetophenone and 3-propoxy4-methoxyacetophenone 1.0g(4.8 mmol) were added to the reaction bottle successively and stir until completely dissolved (5 min); Anhydrous aluminum trichloride was added to 1.4g(10.5 mmol) and reacted at 35℃ for 4 h. The aqueous phase was extracted by dichloromethane (2x30 mL) into 1 mol-L-1 ammonium chloride solution (25 mL), and the extraction solution was combined with spin evaporation and desolubilization, and then separated and purified by silica gel column chromatography [eluent :V(petroleum ether):V(ethyl acetate)=10∶1 to obtain cream-white solid vanilla ethyl ketone.
