About:
α-ketoglutaric acid is an important biomolecule, one of the important intermediates in the tricarboxylic acid cycle, a nitrogen transporter and a symbiotic substance in molecular oxidation. It plays an important role in the metabolism of microbial cells, and is also an important precursor for the synthesis of a variety of amino acids and proteins.

Physiological effects:
α-ketoglutaric acid is a kind of short-chain carboxylic acid with multiple functions, which does not show toxicity and has high stability in aqueous solution. In vivo, AKG is not only an important transitional product in the TCA cycle, but also a precursor of L-glutamic acid, L-glutamine, L-proline and L-arginine. Experimental studies on animals have shown that AKG, glutamine and glutamate ingested by animals are absorbed and metabolized in the upper part of the small intestine, in which up to 95% of glutamate and 70% of glutamine are degraded into CO2, while only 40% of AKG is degraded, and the remaining AKG can be used for the metabolic process of intestinal cells and peripheral tissues.

Preparation:
The steps of bioconversion to synthesize alpha-ketoglutaric acid are as follows:
(1) The bacteria of Max Kluyveria ATCC36534 stored in the refrigerator were inoculated on fresh bevel medium and cultured in a constant temperature incubator at 25 ~ 30℃ for 24 ~ 36h to obtain activated Max Kluyveria ATCC36534 strain. The incline medium consists of glucose 10 ~ 20g/L, peptone 5 ~ 10g/L, yeast leaching powder 3 ~ 5g/L, AGAR 15 ~ 20g/L, water as solvent, natural pH (measured 6.3 ~ 6.5), high pressure steam 121℃ sterilization 15 ~ 20min.
(2) The 2 ~ 3 rings of the slant strains of Max Kluyveromycetes ATCC36534 after activation culture were selected with the inoculation ring step (1) and inoculated into the seed medium. The seed culture after inoculation was based on 20-24h culture at 25 ~ 30℃ and 200 ~ 250r/min oscillating conditions, and the seed solution with a dry cell concentration of 5 ~ 7g/L was obtained. The seed medium composition is the same as the inclined medium in step (1), except without AGAR, and the filling amount in the triangular bottle is 20% ~ 40% of its volume. The triangular bottle is sealed with 8 layers and sterilized by high pressure steam at 121℃ for 15 ~ 20min.
(3) Select the 3 ~ 5 rings of Max Kluyveria ATCC36534 inclined surface bacteria after activation culture in step (1), or the seed solution prepared in step (2), and add 5% ~ 10% of the volume to the transformation medium, and culture at 25 ~ 30℃ and 200 ~ 250r/min for 20 ~ 24h. The dry cell concentration was 6-10g /L, and the final concentration was 10-50mmol /L H2O2, or the final concentration was 0.5-1.0 μmol/L methotrexate, or both were added at the same time. Then L-glutamic acid with a final concentration of 20 to 50g/L was added. Under the same conditions, the synthetic culture medium of α-ketoglutaric acid with the concentration of 17.3 ~ 42.9g/L was obtained in triangle flask for 20 ~ 24h. The composition of the conversion medium is: Sucrose 50 ~ 100g/L, yeast leaching powder 10 ~ 20g/L, KH2PO43 ~ 5g/L, K2HPO45 ~ 6.5g/L, MgSO40.5 ~ 1.0g/L, the solvent is tap water, the pH is natural (measured 6.5), the amount in the triangle bottle is 20% ~ 40% of its volume. The triangular bottle was sealed with 8 layers and sterilized by high pressure steam at 121℃ for 15 ~ 20min.
(4) The synthetic culture medium prepared in step (3) was centrifuged for 3000 ~ 4000g and 5 ~ 10min to remove bacteria, then supernatant a was obtained, activated carbon was added, and activated carbon was filtered after stirring for 30 ~ 60min. The filtrate was reduced pressure and concentrated to 1/2 ~ 1/4 of the original volume, and the sediment was removed again by centrifugation to obtain supernatant b. Add α-ketoglutaric acid seeds, slowly stir and cool to 4℃, stand for 8 ~ 12h, filter and collect α-ketoglutaric acid crystals, vacuum drying at 45℃ to obtain crystalline α-ketoglutaric acid; The volume of the activated carbon added above is 5 ~ 10g/L, and the volume of the α-ketoglutaric acid added above is 10 ~ 20g/L
